ref/PTC-1, -2, and -3 Oncogene Rearrangements in Human Thyroid Carcinomas: Implications for Metastatic Potential?

Sonia L. Sugg, MD, Shereen Ezzat, MD and Sylvia L. Asa, MD, PhD

Mount Sinai Hospital and The Wellesley Hospital Toronto, Ontario M5G 1X5

We are pleased that Mayr et al. were able to extend our findings to another population. They too have demonstrated that ret/PTC expression in papillary thyroid cancer may predict lymphatic metastases in a subgroup of young patients with early tumor stage and otherwise good prognostic features. We agree that ret/PTC-1, 2, 3, and now also -4 rearrangements in papillary thyroid cancer are of biologic significance, but their mechanism of action and possible interactions with other oncogenes or tumor suppressor genes remain to be established. Clearly, the role of these genetic rearrangements in determining the clinical phenotype of thyroid cancer will need to be addressed by further studies utilizing additional methodologic approaches.
