Effect of 17-Ethyl-19-nortestosterone 3-Enol Propionate (SC-7294) on the Metabolism of Adrenal Cortical Steroids

JACK GELLER, M.D.

17-Ethyl-19-nortestosterone 3- enol propionate (SC-7294) administration reduced total urinary neutral 17-KS by interfering with the liver degradation of their adrenal cortical and gonadal precursors. No change in total urinary 17-hydroxy corticoids was noted following this drug. However, it was demonstrated that reduction in ring A of C-21 adrenal cortical steroids by the liver is impaired following the administration of SC-7294 without any significant effect on conjugation of these steroids. This study does not support the hypothesis that SC-7294 reduced urinary neutral 17-KS excretion because of an effect on either pituitary ACTH release or adrenal cortical steroid synthesis.

Supported in part by Research Grants 3838, M2502 and A2965 from the National Institutes of Health and G. D. Searle & Co.
